DNA Fingerprinting Diagnostics (Replication, PCR and RFLP) Kit

Introduce students to the concepts of DNA replication and how it relates to PCR (Polymerase Chain Reaction) and RFLP (Restriction Fragment Length Polymorphism) analysis of DNA—current technology performed by forensic and medical laboratories. Observe how DNA analysis can be used to diagnose disease. Students: • construct specific genes with characteristic nucleotide configuration for cystic fibrosis • observe the difference between normal and mutant genes involved in cystic fibrosis and how the disease can be diagnosed by DNA fingerprinting analysis • use same nucleotides to demonstrate amplification and PCR • simulate gel electrophoresis with “bands of DNA”; simulate analysis of DNA sequences by RFLP • compare and contrast band formation on the “agarose gel electrophoresis” by PCR and RFLP. Basic set with reusable materials can be used by five teams of six students.
